Will a glass straw fit for the lid that has a straw?

Think so, but would depend on what size the glass straw was? If it was too large for the regular straw hole in the lid you could still insert the straw into the sipping/drinking hole.

In order to use the straw, do you have to keep the clear tab in the "up" position?

There is no up or down. It's a slide open and shut. With that said you need to have it open enough for a straw. There is no special spot for a straw.
